海洋Penicillium sp. FJ—1中一个抗真菌活性的新橙酮苷(1)

     [摘要]红树植物内生真菌的代谢物中存在很多新的活性化合物。通过对红树植物海榄雌中一株内生真菌Penicillium sp. FJ-1的研究,利用硅胶柱色谱方法分离得到了一个新橙酮苷(Z)-7,4′-dimethoxy-6-hydroxy-aurone-4-O-β-glucopyranoside(1)。通过波谱分析方法确定其化学结构。化合物1具有显著的抗白色念珠菌活性,强度优于氟康唑,与两性霉素B相当,并且能够浓度依赖性抑制磷脂酶分泌。

    Marine-derived fungi have been proved as a rich source of active secondary metabolites with unique structures and diversified biological activities[1]. Over the last decade, many new metabolites have been isolated and identified from marine-derived fungi, which has been the focus of discovery for new pharmacological products[2]. Mangrove plants are growing in subtropical and tropical intertidal habitats[3]. Many chemical constituents, such as anthraquinones, terpenoids and seiricuprolides were isolated from this plant or mangrove-derived fungus, and exhibited anti-inflammatory, antimicrobial and antiproliferative activities[4-6]. Glycosides, flavonoids and terpenoids are the main constituents in Avicennia marina[7-10]. Preliminary study showed that the EtOH extract from endophytic fungus Penicillium sp. FJ-1 of A. marina exerted inhibitory effect against Candida sp. Further investigation on the metabolites from the culture broth of this fungus resulted in the isolation of a new aurone glycoside (Z)-7,4′-dimethoxy-6-hydroxy-aurone-4-O-β-glucopyranoside (1) (Fig.1) by column chromatography and recrystallization methods. Herein we report the isolation, identification and antifungal activity of this compound.

    1Experimental

    1.1General experimental procedures

    The UV spectrum was obtained using a Shimadzu UV-2401-A spectrophotometer with PhotoMultiplier Tube (Shimadzu, Japan). The NMR spectra were recorded on a Bruker AV-500 spectrometer (Bruker, Swiss), using TMS as an internal standard. The HR-ESI-MS spectra were measured with an API QSTAR Pulsar mass Spectrometer (Bruker, Swiss). Column chromatography was performed with silica gel (200-300 mesh, Qingdao Haiyang Chemical Co., Ltd, Qingdao, China). TLC was carried out on precoated silica gel GF-254 plates (Merck, Germany), and spots were detected with 10% H2SO4 in alcohol followed by heating.

    1.2Plant material

    The plant of A. marina was collected in Zhangpu county, Fujian province, China, in Aug. 2013, and authenticated by Prof. Shang-Jun Yang, Institute of Materia Medica, Shandong Academy of Medical Sciences. A voucher specimen (No. 20130815) was deposited at the department of chemistry of natural product, institute of materia medica, Shandong academy of medical sciences., 百拇医药(宋艳霞 马强 李捷)
